A complete filtration-based purification system for concentrating and purifying infectious adenovirus. The Adeno-X Maxi Purification Kit is an alternative to CsCl density gradient ultracentrifugation. Each kit provides the buffers and filters needed to purify adenovirus from cell lysate.

The Adeno-X Rapid Titer Kit takes advantage of production of viral hexon proteins for the titration of adenoviral stocks in 48 hours. A viral stock is tested by making serial dilutions, infecting HEK 293 cells, and 48 hours later, fixing and staining those cells with the Mouse Anti-Hexon Antibody. The signal is detected with a Rat Anti-Mouse Antibody conjugated to horseradish peroxidase (HRP) and developed with metal-enhanced 3,3'- Diaminobenzidine tetrahydrochloride (DAB).

The Adeno-X qPCR Titration Kit is designed to be used with real-time PCR instruments to rapidly titrate adenoviral stocks from all Ad5-based adenoviral vectors. The protocol combines qPCR with green intercalating dye chemistry, allowing you to determine the viral genome copy number in adenoviral preparations (i.e., crude lysates or purified stocks) from a calibrated DNA standard curve. The kit includes a viral nucleic acid purification kit, a calibrated Adeno-X DNA Control Template, primers, and all the reagents required to conduct 200 TB Green-based qPCR reactions with either the DNA Control Template or purified viral DNA.

Adeno-X GoStix are designed to instantly confirm the presence of adenovirus in cell culture supernatants or lysates. The test detects adenoviral hexon proteins and can be used to determine if infectious virus was produced post-transfection, or for selecting the best time to harvest your virus. The included Hexon Control provides confirmation of GoStix test function.

The Adeno-X 293 Cell Line is a low-passage transformed human embryonic kidney cell line for the production of adenovirus stocks. Adenovirus stocks can be produced by either transfecting this cell line with a linearized adenoviral vector (such as the linear vectors supplied with any Adeno-X System 3) or infecting with a viable adenovirus seed stock. The slower growth rate exhibited by this line reduces cell death due to overconfluence, and therefore facilitates more efficient rescue and amplification. These cells also possess strong adherence and are highly transfectable.

The CAR Receptor Booster increases the transduction efficiency of target cells to infection by adenovirus. The CAR Receptor Booster consists of exosome-like particles coated with adenoviral receptor protein CAR. When applied to target cells, fusion of the particles with the host cell plasma membrane temporarily increases surface levels of CAR to allow for efficient adenoviral transduction. Additionally, CAR Receptor Booster renders cells that cannot usually be transduced with adenovirus (e.g. hematopoietic cells) susceptible to infection.
