Detection of horseradish peroxidase (HRP): Western BLoT chemiluminescent substrate series

The Western BLoT HRP substrate series are chemiluminescent substrates for detecting horseradish peroxidase (HRP)-labeled antibodies used during Western blotting. All of the substrates contain two components: a peroxide reagent and a luminol/enhancer solution. To use, equal volumes of the two solutions are mixed, the blot is incubated with the solution, and chemiluminescence is detected using film or digital imaging equipment. We offer four types of HRP substrates for Western blot detection.

The Western BLoT HRP Substrate series are chemiluminescent substrates for detecting horseradish peroxidase (HRP)-labeled antibodies used during Western blotting. All of the substrates contain two components, a peroxide reagent and a luminol/enhancer solution. To use, equal volumes of the two solutions are mixed, the blot is incubated with the solution, and chemiluminescence is detected using film or digital imaging equipment. We offer four types of HRP substrates for Western blot detection. Please refer to the table in the "More Information" section below to compare the substrates' specifications.

  • The Western BLoT Chemiluminescence HRP Substrate (Cat. #T7101A) provides sensitivity suitable for detection using X-ray film. Although it is the most basic substrate in Western BLoT HRP substrate series, it produces a strong signal and provides high sensitivity with short exposure times.
  • The Western BLoT Quant HRP Substrate (Cat. #T7102A) is intended for Western blot detection using a CCD camera. The signal from Western BLoT Quant is linear with respect to protein amount over a broad range of concentrations. In addition, this substrate produces a strong, long-lived signal, allowing long exposure times to enable the detection of low-abundance proteins. Western BLoT Quant substrate can also be used for X-ray film detection.
  • The Western BLoT Hyper HRP Substrate (Cat. #T7103A) is more sensitive than Western BLoT Quant and is suitable for quantitative detection of low-abundance proteins. Western BLoT Hyper is also appropriate in situations when the amount of primary antibody is limited and a high dilution factor is desired, or when the primary antibody has a relatively low binding affinity. Western BLoT Hyper substrate can also be used for X-ray film detection.
  • The Western BLoT Ultra Sensitive HRP Substrate (Cat. #T7104A) is the most sensitive substrate in the series and is suitable for quantitative detection of very low-abundance proteins. It is possible to detect protein levels in the femtogram range using this product.

T7101A Western BLoT Chemiluminescence HRP Substrate 250 mL USD $303.00

Western BLoT Chemiluminescence HRP substrate is a horseradish peroxidase (HRP) substrate specially developed for X-ray film imaging of Western blots. This product is part of the Western BLoT HRP substrate series and produces a strong signal for high sensitivity with short exposure times. The signal from Western BLoT Chemiluminescence HRP substrate is long-lasting, allowing the blot to be re-imaged. For detection using a CCD imager or for applications requiring higher sensitivity, we recommend using other products in the Western BLoT HRP Substrate series.

T7102A Western BLoT Quant HRP Substrate 100 mL USD $385.00

The Western BLoT Quant HRP Substrate is a horseradish peroxidase (HRP) substrate specially developed for CCD imaging of Western blots. The signal from Western BLoT Quant is linear with respect to protein amount over a broad range of concentrations, allowing full use of the linear range of CCD detection. Western BLoT Quant produces a strong, extremely long-lived signal. These features, in combination with very low background levels, allow for long exposure times to enable the detection of low-abundance proteins. Western BLoT Quant is also compatible with X-ray film detection, although the limited dynamic range of film will make the resulting data less amenable to quantification. For applications requiring even higher sensitivity, we recommend using the Western BLoT Hyper HRP Substrate.

Western BLoT Ultra Sensitive HRP Substrate allows detection of very small amounts of protein during Western blot analysis. This chemiluminescence substrate offers the highest sensitivity, and can be used to detect horseradish peroxidase (HRP)-labeled antibodies bound to target proteins on immunoblots. It is possible to detect protein levels in the femtogram range using this product. This substrate maintains a strong emission signal with linear signal across a wide dynamic range, allowing you to take full advantage of CCD camera detection sensitivity. Since background is very low, this system can be used for long exposure times, enabling detection of very small amounts of target protein. This product is useful when detecting minute amounts of target protein or when a limited amount of primary antibody is available. Western BLoT Ultra Sensitive HRP Substrate can be used for X-ray film detection. However, since the emission intensity is extremely high, it may be necessary to limit the exposure time and adjust the antibody dilution when using X-ray film.
