Easy and efficient lentiviral transduction with a macroporous transduction sponge
In this poster, we described a simple yet elegant product with a reproducible workflow designed to reduce cell touchpoints for a more streamlined experimental design. The Lenti-X Transduction Sponge enables transduction of up to 107 cells per reaction in one well of a 24-well plate. The protocol is gentle to cells, resulting in high cellular viability and preservation of cell phenotypes. No optimization or change of protocol is required for different cell types. The transduction sponge is an excellent alternative to spinoculation and can be applied for transduction applications in any cell type, including hard-to-transduce cells.
Smartphone-based titration of baculoviral and AAV vectors
In this poster, we presented a highly convenient, lateral-flow-based titration technology that can quantify both baculoviral and AAV vector preparations in approximately 10 minutes, reducing expense and production time. The two-step assay analyzes gp64 in viral supernatant. First, the sample is added to a lateral flow device, where test and control bands soon appear. Then, the bands are imaged via a smartphone camera and analyzed using the GoStix Plus app. We show that baculovirus can be detected as early as 4 days post-transfection (P0). Additionally, we were able to screen viral clones (plaques) for titers after only one round of amplification (P1). The resulting titer values were within threefold of current titration methods, with coefficients of variation less than 15%.
