Cell lysis buffer—reagent for protein extraction
xTractor Buffer is the most flexible and efficient cell lysis buffer for protein extraction from bacterial, yeast, mammalian, and baculovirus-infected cells. This cell lysis reagent provides fast and efficient (10 min) lysis using mild, non-denaturing conditions. xTractor Buffer has been optimized for extraction of his-tagged proteins, so it is compatible with all IMAC resins.
xTractor Buffer is the most flexible and efficient cell lysis buffer for protein extraction from bacterial, yeast, mammalian, and baculovirus-infected cells. This cell lysis reagent provides fast and efficient (10 min) lysis using mild, non-denaturing conditions. xTractor Buffer has been optimized for extraction of his-tagged proteins, so it is compatible with all IMAC resins.
The cell lysis method is simple. Just resuspend the cell pellet in the cell lysis buffer (1:20 w/v) and mix gently for 10 minutes, reducing the risk of proteolysis. The final lysate has no visible cell fragments or precipitates. After extraction, simply centrifuge or filter the lysate and load it on any affinity resin column to isolate your tagged proteins. To save time, the resulting lysate can be loaded directly on a TALON CellThru Column without centrifugation or filtration.
This cell lysis protocol is a highly efficient means of extracting proteins from any cells in order to obtain increased protein purity and yield. When compared to sonication, xTractor Buffer extraction yielded more protein content from the same size sample. In addition, the biological activity of the protein obtained by xTractor Buffer extraction is higher than the activity of the protein obtained by sonication.
The xTractor Buffer Kit provides enough buffer to extract proteins from 10 g of bacterial pellet. The buffer kit also includes lysozyme and DNase I. This method works especially well for high molecular weight proteins, such as beta-galactosidase (LacZ), that cannot be extracted unless the membranes are completely disrupted.
Compatible with resin products
xTractor Buffer has been optimized for extraction of tagged proteins, which can then be purified with any of our affinity resins, such as TALON Co Resin or His60 Ni Superflow Resin, or any other affinity resins.
