Magnetic-bead-based isolation of DNA and RNA from water and air samples
By enabling efficient isolation of DNA and RNA from diverse water and air samples, the NucleoMag DNA/RNA Water kit provides a versatile, user-friendly solution for microbial metagenomics and environmental microbiology applications. In addition to allowing for analysis of the entire microbiome—including bacteria, algae, and fungi—concurrent isolation of DNA and RNA can be used to specifically determine the number of living microorganisms in a sample and to measure the efficiency of sterilization procedures.
This kit is compatible with a variety of filters and filtration systems (including 25-mm, 47-mm, and Sterivex cartridge filters) and can be used to process both water (fresh and saltwater) and air samples. The scalable magnetic-bead-based technology employed by the kit enables high-throughput sample processing and allows for the protocol to be performed manually (using magnetic separators) or automatically on popular platforms such as KingFisher and epMotion. Purified DNA and RNA are suitable for downstream applications such as qPCR and NGS.

NucleoMag principle
The NucleoMag family of products is based on magnetic-bead technology. Nucleic acids are bound to the magnetic beads under chaotropic salt conditions, contaminants are washed away with ethanolic wash buffers, and pure RNA and DNA are eluted from the beads using elution buffer.

Various water samples and an air sample were filtered, DNA was extracted with the NucleoMag DNA/RNA Water kit, and the samples were analyzed by qPCR. Microbial DNA was efficiently measured for all of the samples, demonstrating the versatility of the NucleoMag DNA/RNA Water kit.
Figure 4. Compatibility with different homogenization devices. Nucleic acids were extracted from spiked or unspiked tapwater samples with the inclusion of a homogenization step on either a Vortex Genie 2 device (using the MN Bead Tube Holder) or a dedicated instrument (Retsch), and resulting yields were measured by qPCR. Both homogenization approaches yielded comparable results for each sample type, indicating that the NucleoMag DNA/RNA Water kit is compatible with multiple homogenization devices.
Figure 5. Compatibility with different filtration systems. qPCR was performed with nucleic acids isolated from either round filters or a cartridge system using the NucleoMag DNA/RNA Water kit. Comparable CT values were obtained for each prep, demonstrating that the kit can efficiently process samples from many filtration systems.
Figure 6. Efficient removal of PCR inhibitors. Serial dilutions were performed on nucleic acids extracted from three different samples and the dilutions were analyzed by qPCR. The linearity of the inhibition plots indicates the absence of PCR inhibition for each sample.
Figure 7. Automated processing on popular platforms. The NucleoMag DNA/RNA Water kit was used to purify nucleic acids from two different water samples using the KingFisher (magnetic rods) and epMotion (liquid handling) automation platforms, respectively. qPCR analysis of the purified nucleic acids demonstrated comparable yields from each sample, indicating the adaptability of the kit to both automation approaches.
